چکیده

Background Cyclic nucleotide-gated (CNG) channels are members of the superfamily of pore loop cation channels that are activated by the binding of cGMP or cAMP. The channels are important cellular switches which transduce changes in intracellular levels of cyclic nucleotides into changes of the membrane potential and the Ca2+ concentration. CNG channels play a central role in the signal transduction pathways of vision and olfaction. We have studied the physiological significance of a particular member of the CNG channel family, CNGB1. The two splicing variants of CNGB1, termed CNGB1a and CNGB1b, assemble with CNGA1 and CNGA2/4 to form the native tetrameric CNG channels of rod photoreceptors and olfactory neurons, respectively.

An inward current induced by a putative cyclic nucleotide-gated channel in rat cerebellar Purkinje neurons

The roles of cyclic nucleotide-gated (CNG) channels in sensory transduction have long been recognized. More recent studies found that CNG channels are distributed in multiple brain regions involved in memory and learning, including the cortex, hippocampus and cerebellum. These findings suggest that their functions are not limited to sensory perception, but also to neuronal plasticity phenomena,...

Retinal Cyclic Nucleotide-Gated Channels: From Pathophysiology to Therapy

The first step in vision is the absorption of photons by the photopigments in cone and rod photoreceptors. After initial amplification within the phototransduction cascade the signal is translated into an electrical signal by the action of cyclic nucleotide-gated (CNG) channels. CNG channels are ligand-gated ion channels that are activated by the binding of cyclic guanosine monophosphate (cGMP)...

On the multiple roles of the voltage gated sodium channel β1 subunit in genetic diseases

Voltage-gated sodium channels are intrinsic plasma membrane proteins that initiate the action potential in electrically excitable cells. They are composed of a pore-forming α-subunit and associated β-subunits. The β1-subunit was the first accessory subunit to be cloned. It can be important for controlling cell excitability and modulating multiple aspects of sodium channel physiology. Mutations ...

Inherited disorders of voltage-gated sodium channels.

A variety of inherited human disorders affecting skeletal muscle contraction, heart rhythm, and nervous system function have been traced to mutations in genes encoding voltage-gated sodium channels. Clinical severity among these conditions ranges from mild or even latent disease to life-threatening or incapacitating conditions. The sodium channelopathies were among the first recognized ion chan...
